Figure 3: Proteasome activation effects of anthricin and yatein and their chemical structures. Anthricin and yatein adjusted to the concentration that they are contained in juniper berry extract were added to HEK293T cells and their effects to activate proteasome were evaluated. The effects are shown with relative value when juniper berry extract's effect to activate proteasome is set to 100% (n=8, mean±SEM, ***p<0.001, *p<0.05 vs. control by Dunnett's test)
effect to activate proteasome. We discovered that, when juniper berry extract's effect to activate proteasome is set to 100%, anthricin is responsible for approximately 90% and yatein is responsible for the remaining 10% as shown in Figure 3. More in-depth study is required regarding
the relationship between lignans including anthricin and proteasome activation. However, finding new compounds that activate proteasome will contribute to the clarification of the molecular mechanism that controls proteasome activity.
Juniper berry extract's effect to activate autophagy Successful identification of active components led to the discovery of a new effect of juniper berry extract. It has been reported that the anthricin analog causes autophagy activation.10 We, therefore, evaluated the autophagy activation effect of juniper berry extract, anthricin, and yatein. As a result, as shown in Figure 4, all of
them were found to significantly activate autophagy during the period 8 to 24 hours. These results suggest that juniper berry extract
found to activate proteasome also has an effect to activate autophagy and that the extract is expected not only to prevent lipofuscin accumulation but also to degrade it due to these functions.
Juniper berry extract's effects to improve skin L-value and reduce age spots A test was conducted to evaluate whether juniper berry extract's effects to activate proteasome as well as autophagy and to reduce lipofuscin accumulation led to the improvement of L-value
