45-65 having forehead fine lines or wrinkles and declaring lack of firmness on their cheeks. An oily serum which contains live LC was
developed using a selected emollient. This emollient was gentler for LC and presented a higher natural quality compared to a mineral oil. The research team was able to set up an
application routine for the clinical test. The probiotic contained in an oily serum booster at 0.42% was added to an emulsion base to form a mix containing 0.05% of LC probiotic. The efficacy of the probiotic formula was
compared to a mix of oily serum (without the LC probiotic) and the emulsion base (placebo). Skin density was measured with a DUB scanner, a skin echogenic imageing tool that uses the physical properties of ultrasound. After 56 days, the probiotic had significantly
increased the dermis density compared to the baseline (Figure 5) and made the skin appear fuller. This clearly indicates the capacity of the new ingredient to help restructure the fibrous collagen network and help promote collagenous re-densification. In addition, it reduced the forehead wrinkles
by 5% compared to the placebo after 56 days. The effect on forehead wrinkles was measured with a Visia skin analysis system (data not shown).
Postbiolift: Improving skin elasticity During ageing, the components of the

extracellular matrix (ECM), which is the central hub for repair and regeneration of the skin, decrease in terms of quantity and their organization is impaired. Elastin, the main protein responsible for skin elasticity and resilience is decreased and becomes more susceptible to degradation. Another feature of the ageing process of skin is
the appearance of uneven pigmentation and dark spots due to the hyperactivation of melanocytes, altered distribution or change of melanin pigments. Postbiolift (hereafter referred to as LC postbiotic) proved effective in vitro and in vivo to reduce these developments in ageing skin. It improved the mechanical properties
of the skin and thus makes it appear more elastic. Moreover, it smooths the appearance of wrinkles around the eyes and contributes to an even complexion.
In vitro testing The research team evaluated the capacity of the LC postbiotic to enhance elastin synthesis in dermal fibroblast monolayers using Western blot for quantification. The results show that the active ingredient at 0.05% significantly stimulated fibroblast synthesis and secretion of elastin by 28%. The capacity to stimulate the synthesis
of elastin partners by fibroblasts was also assessed. These partners include proteins such as microfibrillar-associated protein 4 or MFAP4,

Fibulin-5 and Elastin microfibril interface- located protein-1 or EMILIN-1. Results showed a significant increase in
MFAP4, Fibulin-5 and Emilin-1 synthesis by 60%, 50% and 61% respectively, which would improve elastic fibre synthesis and assembly to promote a strong and functional elastic fibre network (Figure 6). Assessing the impact of the LC postbiotic on
tyrosinase activity, an enzyme that plays a key role in the melanogenesis process, was also part of the study. Inhibiting its activity can decrease melanin production in the skin.9 The LC postbiotic showed a significant
capacity to limit tyrosinase activity. This effect was confirmed by further tests on melanin synthesis in melanocytes and keratinocytes co-culture using melanin quantification by spectrophotometry. At 0.1%, it decreased melanin synthesis significantly by 32%. Finally, the researchers evaluated the
capacity of the LC postbiotic to lower the frequency of cell contraction in a model of motor neurons and muscle cell co-culture. The cellular contraction was lowered by 82% just two hours after treatment at 0.002%. As the lower frequency of cell contraction leads to skin relaxation, this may contribute to a decrease in the appearance of expression lines.
