protein shell called a cell envelope. Cell envelope is composed primarily of two proteins, loricrin and involucrin. These proteins contain extensive links between each other making the envelope the most insoluble structure of the corneocyte. This structure provides the vital mechanical and water permeability barrier functions necessary for the survival of human being in the terrestrial environment.1
Sensitive Skin/Atopic Skin Sensitive/atopic skin is a complex dermatological condition characterised by an epidermal barrier dysfunction associated to an inflammatory syndrome. Heterogenous and multifactorial, sensitive/atopic skin are the result of the interaction between both environmental and genetic factors: l Mutations of the FLG gene encoding the structural protein, filaggrin represents the most significant factor predisposing to sensitive skin;

l A decrease of the synthesis of lipids lamellae leading to exacerbated breakdown of the epidermal barrier;
l Environmental factors, including the use of soap and detergents exacerbate epidermal barrier breakdown attributed to the elevation of stratum corneum pH which also decreases the activity of the lipid synthesis enzymes.2 Defects in the protective physical barrier lead to a decrease in expression of markers such as filaggrin, involucrin and an overexpression of carbonic anhydrase II. Filaggrin (FLG) is a major structural
protein that binds to keratin filaments and assembles them into macrofibrils. Filaggrin plays a major role in the barrier function. Moreover, the filaggrin degradation into small peptides leads to the formation of Natural Moisturizer Factor (NMF) involved in water retention and regulation of epidermal homeostasis. Involucrin (INV) is the first protein identified as a cornified envelope constituent and plays an important role in the structuring of the corneocytes and the
formation of the cornified envelope. Carbonic anhydrase II (CA2) is a member of the family of metalloenzymes and is involved in maintenance of cellular pH, water transport, and ion homeostasis and increases in atopic skin such as atopic dermatitis. Furthermore, inflammatory syndrome generates morphological changes characterised by intraepidermal edema called spongiosis which are an abnormal accumulation of fluid in epidermis. In subacute stage, pyktonic nuclei of inflammatory cells are observed (Fig 4).
In vitro test and results
The objective of the study is to analyse the positive effects of Natura-Tec Marine OceanMist CL (now referred to as ‘the new active’) on in vitro 3D model of sensitive/atopic skin (AS). The study is conducted on a Reconstructed Human Epidermis (RHE) stimulated by a Th2 cytokine cocktail mimicking a Th2 driven inflammation (RHE/Th2) as observed in atopic dermatitis.
