Results SCBE upregulates SIRT6 and FOXO3 gene expression To determine the capacity of SCBE to increase the expression of the longevity genes SIRT6 and FOXO3, NHEK were treated with SCBE for 24 hours. As shown in Figure 2, SCBE significantly increased SIRT6 expression by 181% compared to untreated cells. FOXO3 expression increased by 104% in


keratinocytes treated with SCBE. Thus, SCBE significantly increases the expression of these two longevity genes, which themselves control various cellular mechanisms involved in healthy skin life expectancy.


SCBE upregulates LC3 and LAMP2 gene expression and protein synthesis In order to determine the ability of SCBE to stimulate gene and protein expression of these autophagy markers, NHEK were treated with SCBE for 24 hours. 44% respectively versus control. These results were then validated at protein


level, as SCBE increased protein detection of LAMP2 and LC3 by 25% (Figure 3B) and 26% (Figure 4B) respectively. Schisandrin alone applied as a positive control resulted in a significant increase of 18% in LAMP2 and 19% in LC3: it therefore participates in the activity of SCBE on autophagy.


SCBE upregulates mitochondria activity Mitochondrial activity from NHEK was measured using a fluorescent probe (MitoTracker®) capable of specifically penetrating active mitochondria. Fluorescence, proportional to mitochondrial activity, increased by 55% with SCBE and shisandrin alone as compared to the untreated control (Figure 5), demonstrating the involvement of this molecule in the positive action of SCBE on mitochondrial activity.


SCBE increases skin dynamic elasticity As described in the Materials and methods section, the elastic deformation of the face was measured at D0 and D28 after daily application of SCBE 1% and placebo in a split-

face manner. An air blow was applied on the lower part of the face, and the recovery time of the skin was measured in milliseconds thanks to a high-speed camera. The faster the skin returns to its original


states, the more elastic it is. After 28 days of application, as shown in


Figure 6, SCBE1% induced a 16.8% decrease regarding recovery time after blowing the lower part of the face. The air blown experiment shows that SCBE1% significantly increases skin elasticity and prevents sagging.


SCBE reduces existing wrinkles After daily application of SCBE 1% during 28 days, wrinkle number was counted on crow’s feet area with an AEVA-HE 3D measurement technic at D0, D14 and D28. SCBE shows a 4.1% decrease in skin roughness and a 10.8% decrease in wrinkle counting compared to placebo (Figure 7).
