Method Fibroblasts of a 39 year-old female volunteer were
seeded into six-well plates and incubated in a CO2 incubator (37°C, 5% CO2
) overnight. When the cell
laying rate in the six-well plate reached 50–60%, test substances were added to the wells (control (0%), 0.1%, for each concentration: N = 3). Then the six-well plate was further incubated (37°C, 5% CO2
), for 24 hours. After incubation, the culture medium was
discarded, and the cells were washed twice with PBS. Subsequently, 1 ml AG RNAex Pro Reagent was added to each well. After RNA was extracted and reverse-transcribed into cDNA, fluorescence quantitative PCR was performed, and the results were calculated. GraphPad Prism software was used to
analyse the data, and the result was expressed as Mean ± SD. The comparisons between groups were performed using t-test statistical analysis. Statistical analysis was two-tailed. p <0.05 was considered as significant difference, p <0.01 was considered as extremely significant difference.
Results For each of the MMPs, the treatment with SC Complex led to a reduced expression (Figures 5, 6 and 7). SC Complex shows to have a protective effect against the breakdown of a large number of dermal compounds, which indicates that it protects against the loss of skin firmness and elasticity and the formation of wrinkles.

To obtain more information on the real- life effectiveness of SkinCharge CLR, further studies were performed on human skin explants where SC Complex was applied topically.
Method Human skin explants were transferred into a six-well plate, and 3.7 ml culture solution was added to each well for further culture at 37°C at 5% CO2
, where the
culture solution was changed every day for two days. After these two days, the skin explants were irradiated with 30 J/cm2

Then the cell culture medium was refreshed, after which SC Complex (3%) was applied topically on the skin explant. Subsequently the skin explants were incubated for 24 hours at 37°C and 5% CO2
. The above-described cycle of irradiation,
refreshing of the culture medium, and the topical application of SC Complex was performed a total of five times. Then the skin explants were further treated topically with SC Complex for another three days, where it was applied on the skin explants

every day. Afterwards, the skin explants were taken for detection and fixed with 4% paraformaldehyde. After 24 hours of fixation, immunofluorescence
detection was performed and pictures were taken under a microscope, with images being collected and analysed. Results It could be shown that the topical treatment of human skin explants with SC Complex is associated with a clearly higher presence of fibulin-5, fibrillin-1, elastin, collagen I, collagen III and collagen XVII (Figure 8). These results convincingly show that the treatment of skin with SC Complex indeed empowers the skin to produce dermal compounds which are indispensable for the maintenance of skin firmness and elasticity.
