and 7% glycerol. The luciferase activity was then measured using an Autolumat LB 9501 (Berthold Technologies, Bad Wildbad, Germany) following the instructions of the luciferase assay kit (Promega, Madison, WI, USA). The background obtained with the lysis


buffer was subtracted in each experiment, and the specific transactivation was expressed as a percentage of Forskolin stimulation (the latter was set to 100%).


Results At two different concentrations, treatment with the cherimoya active showed a strong agonistic effect on the CB2 receptor, where the higher concentration of the two (0.05%) was even able to outperform the positive control, a specific agonist for CB2 (Figure 1).


In vivo study Perception of skin health and wellbeing Volunteers (n = 22) who applied the cherimoya active (3% in an aqueous gel) were given a questionnaire after 17 days of twice daily application.


Assessment of skin sensitivity One application of test product on 17 (for 250 Hz) and 18 (for 5 Hz) volunteers - including five with atopic dermatitis, three with type IV allergy, six with sensitive skin, and one with diabetes type II. Skin sensitivity was determined by measuring current perception threshold (CPT) with Neurometer CPT/C (Neurotron Inc., Baltimore, USA) at 250 Hz and 5 Hz before and 30 minutes after application of the test product. Baseline at t=0 is set at 0%. Values are based on averages of individual results of all volunteers.

Results The 250 Hz experiment showed that the formulation with the cherimoya active outperformed the corresponding placebo formulation by more than 35%. The 5 Hz experiment showed even clearer results: the formulation with the cherimoya active performed more than 60% better than the placebo formulation (Figure 2). These results show that the cherimoya active clearly makes skin less sensitive.

Results In this last study consumers were questioned on whether a simple aqueous gel, showing minimum sensory and biological properties, containing 3% cherimoya active led to improvement of skin health and wellbeing. After 17 days of daily use of the product, the


consumers found that both the intensity and the frequency of itching were reduced by almost 73%. A total of 54.5% of the users confirmed that the feeling of tension in their skin – a phenomenon often encountered by people with dry and unhealthy skin – was perceivably reduced.


The same number of users reported that the


quality of their life was improved, and almost 64% of test persons stated that using this product had improved their wellbeing after only 17 days (Figure 3).


Conclusion Effectively addressing a consumer need implies


understanding its psychology and physiology. When it comes to the topic of ‘health and wellbeing’, combined with the care of skin, it is simply not possible to fully understand every aspect of this topic. What does health and wellbeing mean


for the consumer? What does skin health mean from a scientific point of view? How is this linked with emotional wellbeing? Many questions with many holistic and incomplete answers. To provide an effective solution, zooming in on the most important currently known cell biological features of skin health in the context of emotional wellbeing. What is also required is thorough understanding of how these cell biological features interact with each other. CLR identified cell biological features TRPV1,


CB2 and AC and gained full understanding of the interaction between these features within the endocannabinoid and endovanilloid immunological subsystems inside the skin. An in vitro study described in this article


shows that CLR’s cherimoya active acts as an agonist for CB2. As explained above, this activity intelligently leads to the reduction of activity of TRPV1. Studies not shown in this article confirm that the cherimoya active indeed strongly and specifically reduces TRPV1-induced inflammatory processes. The cherimoya active agonistic effect on CB2


was shown in in vivo studies to strongly and particularly quickly reduce sensitivity of skin. Indeed, a link could be shown between the cherimoya active’s local activity in the skin and overall emotional wellbeing as well. A clear majority of the volunteers taking part in the studies, reported that even a simple gel with 3% cherimoya active improved their quality of life and wellbeing.
