profile across multiple skin-ageing and barrier- related endpoints. After 14 days of topical use, subjects showed improvements in stratum corneum hydration, wrinkle-related parameters, firmness and elasticity, alongside lower redness and reduced transepidermal water loss. At 28 days, the improvements became more


pronounced: stratum corneum moisture increased by 49.45%, wrinkle parameter SEw decreased by 30.02%, redness area decreased by 61.10%, TEWL decreased by 23.57%, skin firmness improved by 45.62% and elasticity parameter R2 increased by 30.28% relative to baseline. These outcomes suggest that the ingredient does not act only as a short-lived sensory tensor. Instead, it appears to influence hydration, barrier function and visible wrinkle endpoints in parallel with elasticity-related metrics. From our perspective, that pattern is coherent.


The β-spiral structure is central to this effect. As the key secondary-structural feature underlying the recoil and resilience of native elastin, it is this spring-like β-spiral architecture that gives elastin its functional mechanical behaviour. If a recombinant humanized elastin can successfully biomimic this structure while remaining formulation-stable and skin-accessible, its benefits should not be limited to firming language alone. By reproducing the β-spiral conformation,


the material is designed to support tissue recoil, improve the recovery of fine lines after facial movement, and contribute to a more resilient extracellular environment. This may also help explain concurrent improvements in barrier-related parameters and visible redness. In this sense, recombinant humanized elastin represents an attempt to translate the functional significance of the β-spiral structure into a usable topical anti- ageing platform.6


Why the two technologies are complementary The most interesting aspect of these two biomaterials is not merely that both are

recombinant. It is that they target different bottlenecks in skin ageing and repair. From our perspective, recombinant PDRN is functionally upstream: it supports recovery signalling, helps moderate inflammatory burden and contributes to the biochemical environment required for fibroblast activity and matrix rebuilding.1-3


Recombinant


humanized elastin is functionally downstream and structural: it addresses the physical architecture that determines recoil, deformation recovery and the visible manifestation of laxity.4,5 This distinction matters in formulation strategy.


Many anti-ageing products improve hydration and perhaps stimulate collagen-associated pathways, yet they do little to address the mechanical deficit caused by elastin loss. Conversely, a structurally focused material may perform better if the surrounding tissue environment is not dominated by inflammation and repair insufficiency. Combining recombinant PDRN with recombinant elastin therefore offers a plausible ‘two-level regeneration’ model: one ingredient supports the repair programme, while the other contributes to rebuilding the tissue mechanics that make rejuvenation visible. Such an approach is particularly suitable for products positioned around fragile, tired,


post-stress or peri-procedural skin, as well as premium daily anti-ageing care. In serum systems, recombinant PDRN could contribute rapid soothing and repair-support benefits, while recombinant elastin could reinforce longer- horizon firmness and wrinkle claims. In masks, eye patches and emulsion systems, the pairing may also fit well with humectants, barrier lipids, niacinamide, ectoin or hyaluronate, depending on the desired balance between comfort, regeneration and elasticity.


A sustainability and platform perspective The other important implication of these materials is manufacturing philosophy. Animal- free recombinant production does more than improve storytelling. It helps address the practical issues that increasingly shape ingredient adoption: secure supply, batch consistency, traceability and compatibility with global sustainability expectations. For recombinant PDRN in particular,


recombinant production removes dependence on large-scale extraction from marine biological material while making sequence engineering and industrial scalability more realistic. For
