Treatment with 2% Exosome-rich extract
for 24h resulted in an increase in mitochondrial mass by approximately 1.39-fold compared to the untreated cells. This rapid increase in mitochondrial mass indicates immediate boost in cellular energy levels, demonstrating the ingredient’s potential for high-performance cosmetic formulations that deliver noticeable results in a short period of use.2
Boosting lysosomal activity, enhancing natural detoxification, radiance and preventing breakouts Lysosomes — often referred to as the cell’s ‘recycling centres’ — play a critical role in breaking down waste, damaged components and toxins to maintain cellular health. As cells grow older, lysosomes can experience alterations that reduce their effectiveness in preserving cellular health.8 BJ cells were treated with 2% Myconoside-rich extract and 2% Exosome-rich extract for 72 hours. Lysosomal activity was then assessed using
neutral red dye staining. Both qualitative and quantitative analyses were performed with image analysis carried out using ImageJ software. Results
A CONTROL
showed a positive effect on cellular detoxification, with a 1.33-fold and 1.28-fold increase in lysosomal mass, respectively. Enhanced cellular detoxification helps boost skin radiance, reduce inflammation, promote anti-ageing effects, improve the absorption of active ingredients and prevent breakouts.2
Upregulating the expression of collagen genes Collagen production, cell cycle regulation and oxidative stress defence are key processes that maintain skin structure, resilience and youthful function. To demonstrate the impact of the Myconoside-rich extract and the Exosome-rich extract on these vital pathways, gene expression analysis was performed for COL1A1, COL3A1, CDKN1A (p21) and NFE2L2 (NRF2). These genes were selected due to their central
roles in maintaining skin firmness, elasticity and resilience—features that naturally decline with age. Enhancing collagen production supports the extracellular matrix, while activating antioxidant pathways and reducing senescence markers helps protect skin cells from ageing-related damage.
2% MYCONOSIDE-RICH EXTRACT - 24h
By targeting these pathways, the Myconoside- rich extract and the Exosome-rich extract show strong promise for use in anti-ageing and skin- rejuvenating cosmetic formulations.2 Total RNA was extracted from treated and
control samples using a purification kit with DNAse I treatment to eliminate DNA contamination. Then, 280 ng of RNA was reverse-transcribed into cDNA. Quantitative real-time PCR was performed using specific primers for target genes (COL1A1, COL3A1, NFE2L2, CDKN1A) and the reference gene ACTB. Gene expression levels were analysed using the delta-delta Ct method, with changes considered significant at a twofold difference or higher.2
COL1A1 (collagen type I) И COL3A1 (collagen type III) 2% Myconoside-rich extract upregulated the expression of COL1A1 – 4-fold and COL3A1 – 5-fold. 2% Exosome-rich extract upregulated the expression of COL1A1 – 4-fold and COL3A1 – 4.5- fold. These results indicate increased production of type I and type III collagen, which are essential for maintaining skin elasticity and structure and contribute to skin resilience and hydration— factors that typically decline with age.2
