Recombinant humanized elastin: addressing the mechanical dimension of ageing The recombinant humanized elastin platform we review here addresses a more structural aspect of skin ageing. Elastin is essential to the formation of elastic fibres, which allow skin to stretch and recover while maintaining tissue integrity over repeated mechanical challenge.4,5 When elastin becomes depleted or


disorganised, the result is not only wrinkling but also sagging, slower recoil and reduced resistance to deformation. Conventional elastin actives have struggled to overcome this because elastin function depends heavily on conformation and supramolecular behaviour.6 Our recombinant humanized elastin is intended


to address the classic barriers that have historically limited the use of elastin: lack of secondary structure, poor water solubility, low expression yield, purification difficulty, endotoxin burden and poor transdermal delivery. The resulting material is described as a recombinant humanized elastin with 100% human-derived functional sequence identity and a biomimetic β-spiral secondary structure intended to reproduce the spring-like behaviour associated with native elastin. This structural claim is central. A primary-sequence elastin mimic is not necessarily enough; functional performance depends heavily on conformation and supramolecular behaviour. According to our technical dossier, the


ingredient is produced through an integrated platform including rational sequence design, secondary-structure folding control, soluble high-expression fermentation, specific enzymatic purification, endotoxin removal, transdermal delivery engineering and signalling-network integration. The reported final material reaches 98% purity, with an endotoxin level of 0.01 EU/ mL, well below the 0.5 EU/mL industry standard. For cosmetic developers, such parameters matter because safety perception and stability are often decisive when introducing biologically sophisticated proteins into leave-on care. Delivery has historically been a major objection

in skin permeability, rising from baseline to 5.62% at 0.5 h, 7.95% at 1 h, 10.62% at 2 h and 17.95% at 4 h, equivalent to a 7.5-fold increase versus 0 h. While proprietary delivery data should always be interpreted in the context of the exact test model, these findings are highly relevant to formulators: elastin is only meaningful as an active if it can reach the skin layers where elasticity is functionally determined. We have also observed notable formulation

robustness. Our dossier states that recombinant humanized elastin maintained activity without detectable degradation across temperatures from -20 °C to 60 °C and across pH 2-9 for as long as 189 days, while also showing compatibility in creams, serums and hydrogel-style applications. This is not a trivial advantage. Many high-value biomolecules fail not because they lack activity, but because they are too unstable for real-world formulation, manufacturing and shelf-life conditions.
