The anti-ageing effects of 2-aza-8-oxohypoxanthine

Vitamin C60 BioResearch Corporation has developed the world’s first cosmetic ingredient containing 2-aza-8- oxohypoxanthine (AOH), under the trade name Repista. This article describes the cellular activation effects of the AOH active ingredient on normal human epidermal keratinocytes. DNA microarray analysis using NHEKs suggested the AOH active ingredient promotes epidermal turnover, differentiation, and maturation, as well as the metabolism of old skin cells. In addition, all tests in the safety evaluation conducted on the AOH active ingredient were negative, indicating that it is a safe cosmetic ingredient.
2-aza-8-oxohypoxanthine (AOH; Figure 1), is a plant growth factor discovered by Honorary Distinguished Professor of Shizuoka University Dr. Kawagishi.1
Until this discovery, there were
only seven plant growth hormones recognized worldwide, making AOH the eighth plant growth hormone to be discovered. We expect the structure of AOH will be recognized as a major discovery and become textbook knowledge. AOH was found to increase rice yields by
approximately 10% in a study by Shizuoka University in Japan (Figure 2A). A cut rose flower in water containing 0.001% AOH remained bloomed for a longer time than flowers in AOH- free water (Figure 2B). Similarly, turfgrass in AOH-free water wilted
faster than that in AOH-supplemented water, once the turfgrass was left to grow without water

replenishment (Figure 3). These results indicate that AOH not only promotes plant growth and increases grain yields, but also contributes to plant homeostasis by prolonging the life of cut flowers and increasing tolerance to drought stress. Research at Shizuoka University has shown
that AOH is a natural, safe compound found in crops, such as wheat, rice, corn, potatoes, tomatoes, truffles and so on.2
Considering the
growth-promoting effect of AOH on plants, Vitamin C60 BioResearch Corporation (VC60) hypothesized that AOH could be beneficial for human skin, and started joint research with Shizuoka University to applicate AOH as a cosmetic ingredient from 2018. The AOH content in edible plants is
extremely low. For example, white rice contained approximately 3 ng/g of AOH. In other words,

300–400 tons of rice are required to obtain 1g of AOH. Since rice is sold in Japan at approximately ¥1000 yen ($7) per kilo, the cost of extracting 1g of AOH from rice alone is ¥300–400 million ($2m-$2.7m). Consequently, it is futile to extract AOH from
grains and vegetables for practical use. Therefore, we established a method to produce AOH utilizing enzymes secreted by microorganisms, then succeeded in making it water soluble so it can be easily incorporated into cosmetic products.

What is Repista? Repista, hereafter known as the AOH active ingredient, is a water soluble cosmetic ingredient containing 1% AOH (Figure 1). The product information for the AOH active ingredient is shown in Table 1. As a new cosmetic ingredient, the AOH in the
