deeper soft tissues, including adipose, muscle and ligament-support compartments, influence the three-dimensional architecture of the eye contour.1,2,6–10
obtained after the treatment with AI-designed peptide is shown in Figure 5.
Clinical translation: comprehensive eye contour improvement The mechanistic findings were translated into visible benefits in a multicentric, double-blind, placebo-controlled, half-face clinical study involving 118 evaluable volunteers from Spain and China. The panel included both men and women aged 31–73 years, with skin phototypes II–IV and visible eye contour concerns. Volunteers applied a formulation containing
2% of the AI-designed peptide solution on one hemiface and placebo on the other hemiface twice daily for 28 days. Evaluation time points and imaging methods were selected to capture the main visible concerns of the eye contour, including vascular dark circles, eye bags, crow’s feet wrinkles and upper eyelid drooping. After seven days, redness in the infraorbital
area, measured through the a* parameter, decreased by 2.68%, indicating an early improvement in the vascular component of dark circles (Figure 6). In parallel, after 28 days, dark circle pigmentation decreased by 12.24%, while skin lightness increased by 2.94% (data not shown). Eye bags were evaluated by three-dimensional
image analysis. After 28 days, eye bag volume decreased by 8.42% and eye bag area decreased by 6.40% versus baseline (Figure 7). Crow’s feet wrinkle volume decreased by 8.87% after seven days and by 12.26% after 28 days (Figure 8A). Upper eyelid appearance was assessed
using computer vision analysis of eye height as a visible parameter associated with droopy eyelid improvement. After seven days, an increase in eye height was observed, with the selected visual material highlighting a 20.0% difference versus placebo (Figure 8B). These findings are consistent with the in vitro effects observed in muscle models.

Figure 6: AI-designed peptide improves both vascular and pigmented components of dark circles in a clinical study. (A) Representative eye contour images showing the reduction of infraorbital skin redness at baseline and after seven days of treatment. (B) Representative macrophotographs of the eye contour area at baseline and after 28 days of treatment, illustrating the visible improvement in dark circle appearance. (C) Three-dimensional colour map generated from the average pigmentation data of all volunteers after 28 days of treatment, showing a reduction in periorbital pigmentation compared with baseline
Discussion The data presented here support a different framework for understanding periocular ageing. The AI-designed peptide was initially selected for its ability to modulate PAR-1, an upstream regulator of endothelial permeability and inflammation. Through its involvement in vascular and lymphatic barrier dysfunction and NF-κB-mediated inflammatory signalling, PAR-1 may contribute to the amplification of microenvironmental disruption across multiple periocular tissues. The broad efficacy profile observed across
seven soft tissues suggests that targeting this upstream driver may help interrupt the Amplifying Release Cycle and reduce the downstream consequences associated with visible ageing.
The observed effects on barrier function,
pigmentation, extracellular matrix organization, adipose remodelling, muscle function and ligament-support structures support a broad activity profile across periocular tissues. Together, these findings suggest that a seven-
soft-tissue strategy may provide a more complete scientific rationale for holistic eye contour rejuvenation than traditional single-concern approaches.
Conclusions Eyeseryl All-In peptide represents a new- generation approach to eye contour care, in which artificial intelligence-assisted discovery was used to identify a peptide with activity against a central regulator of the periocular microenvironment.
