The SEM image shows amyloplasts from a high moisture content (72 – 82%) potato tuber (Solanum tuberosum). Prepared using the Quorum PP3010 cryo-preparation chamber. Preparation involved cryo-immobilization at -207 °C in slush nitrogen, fracturing at -140 °C, and sputtering with 4 – 6 nm of Iridium. A sublimation period of 6 min. was used for the tuber specimen to reveal both the amyloplasts and cellulose cell wall compartments. Imaged on Tescan Amber.


The SEM image shows pollen from a pumpkin stamen prepared using the PP3010 preparation chamber. A section of stamen was taken and mounted using mounting media (50:50 mixture of Tissue-Tek OCT (Optimal Cutting Temperature) compound and colloidal graphite), vitrified in slushed nitrogen (LN2) at 207 °C, fracturing at -140 °C, sublimed to -90°C for 2 min and sputter coated with 4– 7 nm of Iridium. Imaged on Tescan Amber.


The micrograph shown here is of Hydrated Sodium Polyacrylate Hydrogel. The sample itself was sourced from dyed sodium polyacrylate hydrogel beads. The bead was cut into a hemisphere using an exacto knife and mounted using a mixture of tissue tek and colloidal graphite (70:30 ratio) onto a cryo stub. The sample was then plunged freezed in LN2, fractured, sublimated and coated with 5 nm of Au/Pd using the PP3010 cryo system. This sample was then loaded onto a Quorum cold stage and imaged within a Zeiss Evo. A beam energy of 6.00kV was used with a probe current of 16 pA.
