Fast and Simple gDNA Isolation from FFPE tissue

The new MagSi-DNA FFPE kit from AMSBIO facilitates the rapid and effortless extraction of genomic DNA (gDNA) from formalin-fixed paraffin-embedded (FFPE) tissues or cells of mammals.

Utilising a magnetic bead technology exclusive to its development, the MagSi-DNA FFPE kit is applicable for both manual and automated retrieval of gDNA from mammalian FFPE tissues or cell specimens.

The traditional phenol/chloroform extraction or alcohol precipitation procedures, as well as repetitive centrifugation, vacuum filtration, or column separations, are not necessary when using the kit. This reduces the preparation time for 96 samples to a mere 40 minutes after paraffin removal and lysis incubation. The kit is structured to ensure secure handling of potentially infectious samples and avoid cross- contamination between samples.

The MagSi-DNA FFPE kit is fully compatible with the majority of commercial liquid handling robots, and consistently produces high-quality genomic DNA yields. The obtained DNA exhibits a high level of purity and can be directly employed for various downstream applications, such as PCR, NGS, and enzymatic reactions.

The MagSi-DNA FFPE kit can isolate and potentially co-purify RNA, depending on the nature of the sample materials. If co-purification is desired, an extra RNase treatment step must be incorporated into the purification protocol.

Cell Imager with Automated Evidence of Clonality at Day 0

Advanced Instruments is proud to announce the launch of Cell Metric X, a revolutionary new image-based system for high-throughput cell line development. Combining the same high-contrast, high-resolution imaging as its predecessor with the added advantage of automated evidence of clonality (AEC), Cell Metric X enables scientists to quickly identify clonally derived cells at day 0. This reduces the time and effort spent on image analysis needed to confirm successful seeding, and in turn, delivers rapid assurance of clonality with image-based evidence required for FDA IND submission delivered weeks earlier.

Cell Metric X retains many core features, including high quality, high contrast, brightfield imaging; high-throughput 10-position plate stacker; robotic access via flexible API and barcode reader; edge-of-well imaging for optimal single-cell detection, and compatibility with the VIPS® PRO for double lock of monoclonality assurance.

New features include automated Evidence of Clonality, which saves the user time by pre-identifying cell locations, and STUDIUS™ integration for streamlined workflow and quality data management across the Solentim Ecosystem. Additionally, the system offers walk-away automation for reduced hands-on time for high-throughput use, easy integration with third-party robotics, an automatic clonality report for confident submissions, Confluence Precision Scan accuracy within 5%, and automated setup for ease-of-use and reduced learning curve.

Cell Metric X is available either as a single plate system or with an automated, temperature-controlled 10-plate stacker. It’s also fully compatible with third- party robotic integration and is part of the Solentim Ecosystem for cell line development, which includes VIPS® PRO, ICON™, and STUDIUS™. Seamless integration between all products allows for instant triaging of clones without spreadsheet comparison, as well as secure clone tracking and automatically created clonality reports for regulatory submission.

All-Purpose UV Spectrophotometer for Both Batch and Continuous Flow Analysis

Uniqsis has announced a record-breaking demand for its Flow-UV inline UV-Visible spectrophotometer, which has been widely adopted for its compact design and versatility in various industries, including biological, biotech, drug discovery, and industrial applications. The Flow-UV boasts a high-resolution CCD array detector that eliminates the need for calibration or routine maintenance and a Xenon flash lamp source with a lifetime of up to 10 years.

Steve Evans, Commercial Director for Uniqsis, commented: “During the last 12 months, one biotech customer ordered 6 Flow-UV units for nucleic acid purity assessment. In a rapidly growing number of pharma labs the unit is being used for high throughput screening of UV-active drug substances and real time determination of the steady state concentration of products being produced in drug discovery reaction optimisation studies. Researchers are also using the Flow-UV for monitoring algal growth, looking at variations of different metal ion complex solutions in rock cores and optimising microporous membrane production to name but a few of the wide array of applications this versatile spectrophotometer was selected for.”

He added: “The Flow-UV uses fibre optic waveguides that connect the flow cell to the detector, allowing the flow cell to be positioned virtually anywhere in the flow path. To assure linearity of response, up to 5 wavelengths can be selected to monitor the progress of a reaction or monitor concentration changes in a wide variety of media. The Flow UV is supplied with 2 software applications for both flow (dynamic) and batch processes. Customers really appreciate the intuitive method set up and simple data acquisition interface.”
