78 Combination Microplate Reader and Imaging System Launched


BioTek continues providing unique solutions and technological advancements in microplate instrumentation by proudly announcing the first combination Hybrid multi-mode microplate reader and imaging system, the Cytation™3 Cell Imaging Multi-Mode Reader. By combining multi-detection and automated digital microscopy in one instrument, cell biology researchers can now glean data-rich quantitative and qualitative information from their cells like never before. Modular architecture allows users to select only what they need now, and upgrade with additional modes as their needs evolve.


The automated digital fluorescence microscope features brightfield imaging and colour switching through onboard filter cubes. Red (Texas Red), green (GFP) and blue (DAPI) cubes are standard, with a fourth position available. Automated XY stage, autofocus, autoexposure and software features increase throughput of CCD-based image capture, cell counting and other tasks that are tedious when performed with manual microscopy systems. Onboard objectives in a magnification range from 2x to 20x allow the user to view an entire microplate well or to examine the minute details of intracellular activities.


Cytation3’s multi-mode microplate reader measures absorbance, fluorescence and luminescence detection. BioTek’s patented Hybrid Technology™ combines filter- and monochromator-based fluorescence optics in one compact unit for endless assay flexibility now and in the future. Filter-based optics provide superior sensitivity and efficiency, while monochromator optics allow any wavelength choice from the UV to the near infrared, as well as wavelength scanning. Top and bottom optics enhance Cytation3’s flexibility even further.


In addition to imaging and reading, Cytation3 offers uniform temperature control to 45°C,


variable shaking modes, and CO2 and O2 regulation for live-cell assays. This streamlines lab efficiency and reduces cell exposure to unregulated environments normally occurring during manual plate transfers.


Cytation3 is ideal for microplates from 6 to 384 wells and microscope slides. The integrated Gen5™ Data Analysis Software is simple and intuitive to use, and requires minimal training. Optional dual reagent dispensers may be used for inject-and-read assays. Heating Stage used to Study Liquid Crystals at the University of the Orange Free State


Researchers in the physical chemistry department of the University of the Orange Free State (UFS) have examined the discotic liquid crystal properties of a number of long chain metal free and zinc phthalocyanines using the Linkam THMS600 hot stage.


The samples were studied using the THMS600 stage in conjunction with an Olympus BH-2 polarising microscope. The scientists were able to determine the existence of the different phases by comparing the observed texture with data from previous X-ray studies. As many as three mesophases were detected within the non-metal and zinc derivative compounds as they were heated. The scientists found changes in observable textures were a function of the heating/cooling rates used; for example when the C15 metal free derivative was cooled at 10°C min-1 was a needle like texture while slow cooling at 2°C min-1


the result resulted in a fan-like texture.


Dr Eleanor Fourie, a lecturer in the department, commented that they had been using the system for over seven years for "a variety of applications including organometallic synthesis and associated studies. The microscope and Linkam hot stage are used to test the liquid crystal properties of some phthalocyanines. It is also used for basic melting point determinations." These compounds were of specific interest as some phthalocyanines within their mesophase have shown rapid response and recovery time to gas exposure and the zinc phthalocyanines in particular have demonstrated beneficial properties for cancer photodynamic therapy.


The properties of each compound are dependent on chain-length and metal constituent, and with the large number of variants to study, the research on synthesis, properties and industrial applications is an ongoing project. As they are understood better, these highly useful chemicals become relevant to old, new and cutting edge scientific applications.


The THMS600 has a wide temperature range of -196°C to 600°C and samples can be quickly characterised by heating to within a few degrees of the required temperature at a rate of up to 150°C min-1 changes closely.
