New Blood Collection Tubes Stabilise Nucleated Blood Cells


The new Streck Cell-Free DNA BCT® CE blood collection tubes, now available in the UK from Alpha Laboratories, contain a preservative that stabilises nucleated blood cells. This unique stabilisation prevents the release of genomic DNA (gDNA), allowing isolation of high-quality cell-free DNA (cfDNA) samples.


These can then be used for a wide range of downstream applications in clinical research studies, drug discovery and diagnostic assay development such as non-invasive prenatal testing (NIPT), liquid biopsies and post transplantation non-invasive graft monitoring. The Cell-Free DNA BCT CE tubes have also been demonstrated to minimise the degradation of circulating tumour cells (CTCs).


Cell-Free DNA BCT CE tubes reduce the need for immediate plasma preparation and CTC processing due to their unique stabilisation properties. CfDNA and gDNA are stable for up to 14 days at 6°C to 37°C, CTCs are stable for up to 7 days, at room temperature. This enables convenient collection, transport and storage whilst maintaining sample integrity. Enhanced stability and recovery of cell-free plasma DNA minimises variability associated with cfDNA sample preparation.


The trusted technology of Cell-Free DNA BCT® CE glass tubes is now available in the Fusion2™ hybrid plastic tube. Fusion2 is composed of a medical-grade plastic with a silicon-based internal barrier coating, providing the benefits of glass with the safety and convenience of plastic.


The new Fusion2 tube offers sample stabilisation identical to the glass Cell-Free DNA BCT CE, which have been used by researchers in studies and clinical trials worldwide. The evaporation of valuable stabilising reagent over time is prevented in Fusion2 tubes leading to uncompromised sample integrity. The tubes are extremely robust and durable, making them resistant to breakage. The silicone-based barrier layer prevents oxygen from permeating through the plastic allowing tube to keep a strong vacuum which provides consistent draw volume. Moreover, the silicone-based barrier layer prevents leachables from migrating into the liquid samples.

Introduction PCR plates from BRAND are designed to support polymerase chain reactions in several ways. The source materials selected are free of PCR inhibitors and the smooth vessel interior minimises the binding of enzymes and nucleic acid to the walls. In addition, the ultra thin-walled PCR plate design facilitates constant, rapid and precise heat transfer leading to convincing yields and short PCR cycle times. Generating the desired PCR product and shielding it from evaporation are decisive elements of a successful PCR.


The innovative self-adhesive press-to-seal sealing fi lm wins over with easy handling; it is not tacky to the touch and provides superior evaporation protection. The fi lm is highly transparent and can be used for measuring the smallest signals during optical measurements like real-time PCR. BRAND PCR plates and the BRAND PCR sealing fi lms form an masterfully tuned system. The surfaces of the PCR plates and the adhesive side of the sealing fi lms are tailored to each other and reach striking results.


Measurement of evaporation losses of different PCR systems


A mixture of water with the cationic dye methylene blue was prepared. In each PCR plate every well was fi lled with 50 µl of the water dye mixture and sealed with adhesive sealing fi lm. The weighed portion of the plates and the sealing fi lms was determined before and after the fi lling of the wells. The roller was used to ensure a fi rm seal. The PCR plates were then put into the thermal cycler Biometra T1 and a PCR run was performed (table 1).


Finally the weighting portion of the PCR plates was examined again.

Conclusion


To obtain successful PCR results it is important to use a harmonising PCR system. The PCR plates have to be securely sealed to preserve the generated PCR products. The adhesive surface of the highly transparent self-adhesive sealing fi lm of BRAND goes hand in hand with the surface of the BRAND PCR plates. The encapsulated, pressure sensitive adhesive keeps the fi lm easy to handle and non- tacky to the touch. After sealing, areas above the sample wells remain adhesive free and do not distort PCR samples. On top the ultra-thin liner and high transparency allows detection of smallest signals during the Real-Time PCR.
