47 Food & Beverage Analysis PCR Detection Method for Flavour Spoilage Yeast Now Available


Bibby Scientific have announced that it has launched a Techne PCR-based method for reliable and highly specific detection of the yeast, Dekkera bruxellensis, which is a major cause of wine spoilage worldwide, causing large economic losses within the global wine industry.


Flavour-spoiling phenolic compounds released from this yeast lead to undesirable aromas, known as ‘Brett’ taints, that are normally associated with aromas of barnyard, burnt plastic, wet animal and horse-sweat. Detection of the yeast through traditional microbiological techniques can be time-consuming, costly and unreliable. In contrast, real-time or quantitative PCR-based detection methods allow for exceptionally rapid and highly specific identification of the yeast.


Testing for the presence of D. bruxellensis can be determined using the Techne Prime Pro 48 qPCR system in conjunction with the Techne qPCR test ‘Dekkera bruxellensis 26S ribosomal RNA’. The Techne qPCR Kit for D. bruxellensis is designed for the in vitro quantification of D. bruxellensis genomes. The kit is designed to enable the broadest detection possible whilst remaining specific to the D. bruxellensis genome. The kit is comprised of primers and probe sequences that have 100% homology with a wide range of D. bruxellensis sequences based on comprehensive bioinformatics analyses.

Personal Touch Added to Wine Microbiology


The Wine Microbiology and Microbial Biotechnology Laboratory at the University of Adelaide has developed a customised solution offering fully automated sampling for yeast fermentations. Based on a Freedom EVO® 200 platform, this system frees researchers from the need to manually aliquot samples day and night for up to three weeks. Dr Tommaso Liccioli Watson, a postdoctoral research fellow in the laboratory, explained: “We use approaches such as directed evolution and mutagenesis to develop novel yeast and bacterial strains to improve the winemaking process. However, these methods are often very time consuming, requiring large-scale screening of hundreds or even thousands of candidate strains, often from heterogeneous populations. Monitoring the fermentation process for large numbers of samples is very laborious, requiring sampling of fermentation vessels every six hours or so over the course of several days to several weeks.”

We were able to integrate our own custom hardware solutions onto the platform with relative ease, building a walkaway fermentation monitoring solution to generate 96-well microplates ready for loading onto our various analytical instruments. Compared to having to be in the lab every few hours, day and night, this is fantastic.”

New Compact Titrators for Food Analysis The titrators from the Ti-Touch family from Metrohm are compact analysers offering the maximum in the class of stand-alone systems for routine analysis in the food industry.


Several different titrimetric determinations are used in food analysis. Apart from acid-base titrations, this also includes precipitation titrations as well as complexometric and redox titrations, such as: citric / tartaric acid in lemonades and soft drinks; chloride determination in meat and cold cooked meats; determining calcium in milk products; sulphur dioxide in wine; water content determination in coffee, confectionery.


The Ti-Touch titrators are suitable for all of the titrations required in the food industry, not only potentiometric determinations but also applications of the Karl Fischer method for water content determination.


The following models are available for selection: 915 KF Ti-Touch; 916 Ti Touch (Food, Salt, Oil); 917 Coulometer.


The Ti-Touch titrators are operated using a touch screen. Comprehensive user administration is available which, among other things, allows up to 14 favourite icons per user to be saved to the start screen which can be touched to start the respective method.


Thanks to the intelligence of the various components, operating errors are virtually impossible. All system components are identified and registered automatically once they are connected; when the titration method is started, not only the titrants and their titers, but also the electrodes and their calibration data are all verified. Plain text messages will be displayed to the user in case of an error.


The instruments have an Ethernet interface which can be used to implement TCP/IP communication. This means that raw data and tamper-proof PDF reports can all be saved directly to the network or can be printed out on the network printer, without an additional PC. The USB interface allows various input and output devices to be connected, e.g. balances, barcode readers, printers, or USB flash drives.
