For your convenience, Aurion has developed Ready-to-Use Incubation Buffer Solutions with long-term stability, that significantly reduce the risk of background.


The main component is AURION BSA-c™, a unique buffer additive that prevents aspecific binding of antibodies and immunodetection reagents to polycationic sites in the specimen, such as histone proteins in the nucleus.


The Ready-to-Use Incubation Buffer Solutions are applicable in all types of immuno(cyto)chemical procedures, not only in ImmunoGold Silver Staining, and also in immunofluorescence and immuno-enymatic techniques in microscopic and macroscopic applications.

Ready-to-Use Incubation Buffer-PBS (EMS Cat. No. 25561)


The ready-to-use Incubation buffer that can be used in all immuno (cyto)chemical methods.


A protein block step is required. This incubation buffer solution is recommended for those specimens and applications were (ultra)structure is important and preservation of structure and/or extraction is not warranted by the specimen preparation method (e.g., for Tokuyasu cryo sections).


Ready-to-Use Incubation Buffer-PBST (EMS Cat. No. 25562)


A ready-to-use Incubation buffer that combines the positive effect of both AURION BSA-cTM and Tween 20®


with


respect to reduction of charge determined and hydrophobic background. This incubation buffer solution suits those applications were preservation of structure is less important. A protein block step prior to the actual incubation may not be required.


Important Note TWEEN-20®


is a detergent. Surface forces may affect/remove existing hydrophobic complexes in/from the specimen and encapsulate them in micelles. When this is the antigen, this will have a negative effect on signal intensity.

AURION Ready-to-Use Incubation Buffer Solutions -PBS and -PBST have a guaranteed shelf life of 18 months from the date of quality control analysis. The buffers should be stored at 4-8°C. Freezing is not recommended

Product Description


AURION BSA-c™ is an acetylated and (partly) linearized form of Bovine Serum Albumin, having an increased net negative charge by acetylation of amino groups of basic amino acids. AURION BSA-c™ will improve signal-to-noise ratios independent of the immunodetection system being used.


TWEEN-20®


is a non-ionic surfactant, a detergent with hydrophobic alkyl side chains. When other hydrophobic components (proteins, antibodies, gold conjugates etc.) are present these components are likely to get embedded in micellar structures. One of the main effects is the suppression of hydrophobic (background) interactions. It acts as a surfactant and thus will enhance reagent spreading in (automated) immuno incubation procedures

AURION ready-to-use incubation buffer solutions are available in two compositions: EMS Cat. No. 25561


PBS (10mM Phosphate buffer, 150mM NaCl) + 0.1% BSA-c™ + 15mM NaN3, pH 7.4. It is recommended to include a protein block step in the immuno incubation procedure (See protocol no. 1 in Technical Tips).


EMS Cat. No. 25562 PBS (10mM Phosphate buffer, 150mM NaCl) + 0.1% BSA-c™ + 0.1% TWEEN-20® AURION ready-to-use buffer solutions are supplied in 1000 ml quantities. +


15mM NaN3, pH 7.4. A protein block step may be omitted from the immuno incubation procedure (See protocol no. 2 in Technical Tips and the Important Note).
