New Method Development Solution: Software Support for Analytical HPLC Method Development and Optimisation
Shimadzu has announced the release of its ‘LabSolutions MD’ software for analytical HPLC method development. In addition to established ‘Method Scouting’ functionality that enables automated, quick and simple column and solvent screening, the new LabSolutions MD method development solution supports ‘Analytical Quality by Design’ (AQbD). With a multifactorial experimental design, only a limited amount of experimental data is required to create a defi ned design space.
Computer simulation and retention modelling within this design space allow for fast, statistically informed decisions on suitable,
robust separation conditions, while reducing the risk of human error. LabSolutions MD, in combination with the Shimadzu LabSolutions chromatography data system, provides data management, evaluation, and intuitive reporting, all in a LabSolutions database, ensuring data integrity.

Four New Products for Enhanced Pyrolysis Analysis
Frontier Laboratories Ltd has launched four new products around the Multishot Pyrolyzer EGA/PY-3030D, their state-of-the-art vertical furnace pyrolyser (for Py-GC/MS), equipped with a programmable ceramic heater, enabling, beside single shot pyrolysis, thermal desorption, heart cut and evolved gas analysis.
The Micro-Jet Cryo-Trap MJT-2030E is an excellent tool for energy effi cient rapid cooling and instant thermal desorption (TD-GC/MS). Trapping of low boiling compounds (CO2
, ethane, etc) is possible due to the liquid nitrogen jet fl ow and cooling temperature of -196ºC. It enables automatic heart cut analysis of gases evolved from Multishot Pyrolyzer. The Selective Sampler SS-2010E is provides Heart-Cutting of desired Peaks and comes with a new Flow Switching System.
Various atmosphere gases (He, Air, etc) can be used. The Multi-Functional Sampler MFS-2015E makes splitless Pyrolysis possible without compromises, enabling 100x improved sensitivity (with LOQs in pg range). The MFS shortens analysis time by backfl ushing, solvent venting is also possible. It is the ideal tool for trace analysis, such as µ- and nano-Plastics, and additives detection. The new Cryogenic Mill IQ MILL-2070 is easy to use and simple to operate. Fast and effi cient high-speed milling is enabled by 3D/8-fi gure crushing ball motion. UHPLC analysis of cyclic peptides under LC/MS compatible conditions
Cyclic peptides are characterised by polypeptide chains that are arranged in a cyclic ring structure. This structure offers various advantages over their linear counterparts: on the one hand cyclic peptides show a higher thermal stability and a higher resistance to digestion. On the other hand, their rigid structure results in a better biological activity since the binding to the target molecule is improved. All these characteristics turn them into highly valuable drug candidates.
Cyclic peptides have various applications; the peptides in this application note, polymyxin B sulphate, daptomycin and bacitracin are used as antibiotics for the treatment of bacterial infections, whereas cyclosporin A is used as an immunosuppressive agent. Other cyclic peptides can also have antifungal or antitumor functionalities.
In YMC’s application note ‘UHPLC separation using column of low hydrophobicity’, the UHPLC analysis of four cyclic peptides is shown. Based on the hydrophobicity of the peptides, a YMC-Triart Bio C4 column was chosen for this application. Since a relatively high concentration of acetonitrile is needed for elution, a C4 modifi cation is a good choice since it provides more acceptable retention times. Since a temperature of 70°C is required, YMC-Triart Bio C4 is a better column choice due to its high temperature stability. Sharp peaks are obtained with a mobile phase containing formic acid, which is commonly used for LC/MS analysis. YMC-Triart Bio C4 shows high sensitivity using these LC/MS compatible conditions.
