11 Improved Yield and Specificity in PCR Amplifications

Thermo Fisher Scientific, Inc announced it offers a premium collection of Thermo Scientific Taq DNA polymerases that are licensed and optimised for use in PCR processes. ThermoPrime Taq DNA Polymerase and Thermo-Start®

Taq DNA Polymerase are designed to provide excellent amplification specificity and

improved product yield. In addition, the high purity of these enzymes ensures their consistent and reliable performance. ThermoPrime Taq DNA Polymerase exhibits enhanced thermal stability at DNA denaturation temperatures, ensuring optimal enzyme activity throughout the thermal cycling process and maximum product yield.

Suitable for the amplification of templates up to 4kb in length, this high performance enzyme is ideal for both standard and long PCR applications. Thermo-Start DNA Polymerase is a chemically modified version of ThermoPrime Taq DNA Polymerase, developed to ensure high specificity in hot-start PCR applications. Unlike other hot-start DNA polymerases, the modification in Thermo-Start DNA Polymerase is extremely stable (requiring heating at 95ºC for 15 minutes for activation) and results in sustained activity of the enzyme over an extended number of cycles with high denaturation temperatures. This eliminates non-specific amplification and increases product yield.

These ultra pure enzymes are also available in convenient master mix formats for quick, easy and reproducible setup of reagents – ideal for high throughput applications. The ThermoPrime and Thermo-Start PCR Master Mixes are also available with ReddyMix™, a unique PCR Buffer that is specially formulated with a red tracker dye to allow direct loading of the PCR product onto gel.

Reactor-Ready Lab Reactor

The patented Radleys Reactor-Ready is an innovative and affordable multi-volume jacketed reaction station for labs involved in chemical synthesis or process development. Simple to use, quick to set-up and incorporating a range of innovative features, Reactor-Ready is designed to boost the productivity of your chemistry. Reactor-Ready’s unique quick-release clamp and quick connect hose couplings allow vessels to be exchanged in minutes with ease and without the use for tools. Reactor-Ready’s I-beam support clamp fits all major brands of overhead stirrer and ensures that the stirrer drive is always in the correct alignment.

It allows the drive to be easily lowered and raised without needing to reposition the clamp. A drop-in stirrer coupling allows the stirrer drive to engage with the stirrer guide below and provides powerful stirring up to 800rpm. The precision engineered PTFE stirrer guide provides excellent stirring and sealing performance with whip-free agitation. The system features wide bore, insulated hoses and vessel connections, which offer maximum flow rate and heating/cooling performance. Integral hose manifolds, attached to the support frame also allow rapid and clean thermofluid drain down during vessel exchange

The system accommodates single and vacuum jacketed borosilicate glass vessels from 250ml to 5 litres. All Reactor-Ready vessel kits feature Radleys’ popular V4 PTFE drain stopcock for operation across the temperature range of -60°C to +190°C, a DN100 polished flat flange with o-ring seal, 5 neck glass lid and Pt100 PTFE temperature probe.

ADVERTORIAL RFM990 Flow Cell Refractometer for Automated Systems

The RFM990 Flow refractometer is a five decimal place (RI) wide range Peltier temperature controlled instrument that has been specifically designed for flow applications. The instrument incorporates a special stand that elevates the instrument to an optimum angle as well as a bubble loop on specific cells, ensuring that sample is presented to the prism without entrapped air and therefore ensuring best performance in the most difficult flow configurations.

Supplied as an instrument module only, the customer can choose from a number of standard cells or for special applications, a custom design chamber may be offered at extra cost depending on viability.

Refractometers and Polarimeters Setting New Standards…

Micro flowcells are typically used in conjunction with an autosampler for use with density meters in breweries for calculating alcohol content and both density meters and polarimeter for flavours & fragrance applications. Micro flowcells are also used in beverage applications, especially in ‘audit laboratories.’

Macro flowcells are used where sample viscosity limits the use of micro-flowcells or for connection to a pilot or small-batch process line, where a normal process refractometer may not be suitable.

Where larger volumes of sample are available, funnel flowcells are often used. These negate the need to clean the instrument prism between sample measurements, providing a rapid sample turnaround, such as in Tare Houses of grape growers co-operatives and sugar mill receiving stations. There are two funnel sizes available.
