IMMUNOPHENOTYPING MULTICOLOUR


OF HEMATOLYMPHOID NEOPLASMS

SUMMARY Multicolour flow cytometric immunophenotyping of hematopoietic cells allows simultaneous visualization of a large number of antigens on a single cell. This in turn provides detailed information on the composite antigenic profile of these cells in health and deviations therein in neoplastic processes. However, realization of limitations in the number of antibody-fluorochrome conjugates that can be concurrently used in an assay tube has shifted the focus on innovative use of bioinformatics software to extract maximum information about the antigenic profile of a cell by summating the data obtained from multicolour flow cytometric analysis. Future research and development in immunophenotyping of hemopoietic cells is likely to follow in this direction. Another noteworthy recent


development in this field is the ability to detect ‘signature’ fusion proteins that result from various chromosomal translocations in neoplastic hemopoietic cells by flow cytometry, thereby obviating the need to perform time consuming and complex cytogenetic and molecular
