Advanced High-Throughput Screening in a microplate reader


H


igh-Throughput Screening (HTS) in microplate readers is especially used in drug discovery and rele-


vant to the pharmaceutical industry as well as academic core facilities. HTS allows researchers to quickly conduct millions of chemical, biochemical or pharmacological tests and establish a basis for drug design. In order to maximise efficiency, screening facilities must constantly strive to increase throughput and minimise sample expendi- ture, while controlling costs. This leads to the key factors in HTS which are highest speed and flexibility without compromis- ing in sensitivity. To meet these requirements BMG Labtech


developed the reference multi-mode microplate PHERAstar® FSX reader for HTS applications. This reader was specifi- cally conceived for the fastest read times, the best sensitivity and unmatched flexibility in all plate formats up to 3456 wells.


Next generation TRF laser The trend towards assay miniaturisation for high-throughput requires the ability to mea- sure the smallest possible quantity. The outstanding sensitivity of the PHERAstar® FSX is based on an innovative optical design which is composed of a free air opti- cal path, three independent light sources, Simultaneous Dual Emission detection and high transmission filters. To ensure best per- formance for any HTS assay the reader comes with a high energy xenon flash lamp, a special laser for Alpha Technology and the next- generation laser for TRF/TR- FRET. The TRF laser with a frequency of 60 flashes per second increases throughput and precision of measurement and allows for a full 1536-well plate to be measured in only 36 seconds (Figure 1).


Simultaneous Dual Emission detection for alpha technology Numerous HTS assays require detection of two emission wavelengths. BMG Labtech pioneered the technique of Simultaneous

Thanks to Simultaneous Dual Emission detection and two pairs of matched Photomultiplier Tubes (PMT) detectors, the PHERAstar® FSX can simul- taneously detect two separate emission wavelengths in one single measurement. This offers a significant speed advantage by cutting read times in half. Moreover, Simultaneous Dual Emission corrects flash- to-flash variations, photobleaching, decay- ing kinetic signals or fluctuating conditions such as temperature, pH and evaporation. Simultaneous Dual Emission detection can be used in any assay that measures two emission wavelengths or polarisation vec- tors, including FP, FRET, HTRF and AlphaPlex.


Best performance in all HTS assays The advantage of miniaturisation allows rapid, inexpensive and effective measure- ment either in 384, 1536 and 3456-well microplates. The PHERAstar® FSX offers a unique combination of features to support all major existing applications and future needs. The reader easily performs applica- tions such as protein-protein interactions, affinity binding assays, compound and inhibitor screening as well as DNA, RNA and protein quantification. Moreover, the microplate reader is suitable for enzyme activity, kinetic, cell-based and reporter gene assays. The PHERAstar® FSX espe- cially provides excellent performance in all HTS applications, including NanoBRET, Alpha Technology and HTRF assays.


Automation in robotic systems Automation is a key factor in HTS, hence microplate readers have to be easily inte- grated in automated processes of modern laboratories. For HTS automation purpos- es, the PHERAstar® FSX offers improved robotic integration capabilities, multi-user control and MARS data analysis software with included digital signature and FDA 21 CFR part 11 compliance, and can be equipped with BMG Labtech’s Stacker. Moreover, data can be exported as Excel or ASCII files.
