PEPTIDES & PROTEINS

Manufacturing deals and technologies for peptide synthesis

The peptides manufacturing sector continues to benefit from new technologies that are enabling supplier companies to fulfil the needs of the pharma and biotech sector as peptide and small protein products progress through the clinic towards full commercialisation. We review some recent manufacturing agreements and technology developments in the sector.

eptide chemistry specialist Bachem and oligonucleotide company BioSpring GmbH have concluded an agreement to co-operate on the manufacture of oligonucleotide-peptide conjugates for R&D projects thus bringing together the required expertise in both oligonucleotide and peptide chemistry from two manufacturers experienced in these fields. Dr Anne-Kathrin Stoller, vice president for marketing and sales at Bachem, comments: “There is an increasing number of examples of new drug development candidates now reaching clinical trials where both types of chemistry are involved. Bachem is delighted to partner with a successful and dynamic company such as BioSpring to offer pharmaceutical and biotech customers an excellent combination of synthetic skills.” “We are particularly enthusiastic regarding this partnership with Bachem” adds Dr Sylvia Wojczewski, CEO of BioSpring. “Both the geographic proximity and the respective technical expertise will help make this partnership productive for both our organisations.”

Bachem is specialised in the process development and the manufacturing of peptides and complex organic molecules as active pharmaceutical ingredients (APIs), as well as of innovative biochemicals for research purposes. The company is headquartered in Bubendorf, Switzerland, and has affiliates in Europe and the USA, and therefore works on a global scale. Headquartered in Frankfurt, Germany, BioSpring is a cGMP-certified company providing synthetic oligonucleotides for therapeutic, diagnostic and research applications. The company´s manufacturing capabilities cover the whole range from small scale for initial research up to commercial quantities. The company also provides a large range of services for analytical development, quality control and characterisation of oligonucleotide-based drug substances and drug products for the pharmaceutical industry.

Dimethylated arginine histone peptides from AnaSpec

AnaSpec, EGT Group is now offering a large selection of Histone GO® catalogue peptides including sequences containing modified

amino acids such as methylated (Me1, Me2 and Me3) lysine; phosphorylated threonine (pThr) or serine (pSer); and deiminated arginine (Cit). The company y is now also offering symmetric and asymmetric dimethylated arginine (Me2s and Me2a) in addition to monomethylated Arginine (Me1). Methylation of arginine residues are mediated by members of the protein arginine methyltransferases (PRMTs) family. PRMT type I adds dimethyl groups asymmetrically to arginine (ADMA), while type II add dimethyl groups (SDMA) symmetrically. Type III adds a monomethyl group to arginine. Type 1 PRMT, co-activator-associated arginine methyltransferase 1 (CARM1), also known as PRMT4, methylates histone 3 at R2, R17 and R26. CARM1 methylation of H3R17 activates transcription. PRMT5, which catalyses H3R8 and H4R3 symmetric dimethylation, leads to repression of gene expression, while PRMT1 asymmetric dimethylation of H4R3 is associated with gene activation.

AnaSpec is the only provider of a long-wavelength HDAC assay kit, the SensoLyte 520 HDAC Activity Assay Kit. The company also provides unusual amino acid building blocks such as Lys(Ac), Lys(Me1-3), Arg(Me2), pSer and pThr for histone peptide synthesis

Arrayjet and JPT Peptide Technologies finalise strategic agreement

German peptide manufacturer JPT Peptide Technologies GmbH has installed a customised version of an Ultra Marathon microarrayer, one of the largest-capacity microarrayers currently available from Arrayjet Ltd.

Bachem has extensive experience in the synthesis of peptides from research quantities to commercial scale.
