Improving silhouette by regulating PGC-1α levels in adipose tissue

Lipid accumulation depends on diet, age, gender and exercise. In women it is usually located in buttocks, thighs and hips, while in men and menopausal women fat mainly accumulates around the abdomen and waist. Adipose tissue is redistributed from the subcutaneous stores to visceral depots when ageing, reducing local volume in some areas (breast or hands) and causing an older appearance. On the contrary, fat deposits can abnormally expand due to several factors such as genetics, imbalance between intake and energy expenditure or a high- calorie diet. This extra growth pushes and distorts the connective tissue, resulting in irregularities in the dermo-hypodermal junction line, local volume increase and cellulite. Therefore, controlling local adipose tissue growth could help to recover the volume lost as a consequence of ageing, and also increase (breast) or decrease (thighs) local volume where desired.


Adipose tissue is a specialised connective tissue strategically located beneath the skin, without anatomic constraints. White adipose tissue (WAT) is


the prevalent type in adults functioning as the major storage site for the lipids from daily intake, which are burned whenever body cells require energy. Adipocytes are the main cells in WAT but this tissue also has preadipocytes (precursor cells, 15-50%), macrophages, fibroblasts, leukocytes and many collagen fibres, which provide support. One of the most relevant features of adipocytes is their capacity to accumulate triglycerides, whilst preadipocytes cannot. Adipocytes contain a single large lipid droplet (80% of the cell content), which forces the nucleus and cytoplasm to remain in the periphery. The enlargement of WAT volume can be caused either by an increase in the number of adipocytes or in lipid content. The differentiation process from preadipocytes to mature adipocytes is a complex process known as adipogenesis, in which Peroxisome Proliferator-Activated Receptor-Gamma


(PPARγ) and Peroxisome proliferator-activated receptor-Gamma Coactivator 1 alpha (PGC-1α) are both indispensable. In WAT, PGC-1α coactivates PPARγ and they positively regulate the necessary gene expression which transforms preadipocytes into mature adipocytes, with lipid storing capacity.


Thus, stimulating or reducing PGC-1α expression would increase or diminish adipocyte maturation and consequently local volume. Lipotec has developed two ingredients for a complete remodelling treatment through PGC- 1α expression in WAT. Silusyne™

assays were performed applying these


ingredients, in order to study its effect in PGC-1α expression and volume in key areas (thigh and breasts respectively). Silusyne™


diminished PGC-1α expression and


lipid accumulation in vitro (even more than caffeine), and also improved irregularities and cellulite appearance in vivo, as a result of the 21% decrease in the length of the dermo-hypodermal junction line. On the other hand, Adifyline™


enhanced PGC-1α


levels and lipid storage in vitro, confirmed in vivo by the volume growth seen in breasts (by 30-fold versus placebo). Considering these results, Silusyne™
