Agarose
Agarose gel electrophoresis has become an indispensable tool in molecular biology laboratories. Agarose is a mixture of complex polysaccharides
extracted from algae. This results in a porous, resilient gel. When subjected to an electrical field, charged molecules will migrate through
the pores of the gel toward oppositely charged electrodes. The rate of migration is determined by the mobility of the charged molecule. The
higher the charge to mass r a t i o of a particle, the higher its electrophoretic mobility. Unlike other separation mediums, the varying siz e of the
agarose pores allows it to be used for separation of almost all classes of biological molecules. Chromosomal DNA exceeding 5 mega-bases
in length or as small as 10 bp can be routinely separated in Agarose gels. Additionally, certain modified Agarose exhibit resolving capabilities
equivalent to that achieved by polyacrylamide. Due to its unique characteristics, Agarose is one of the most widely used mediums in research
and clinical laboratories for the analysis and preparation of biological molecules.
Agarose can vary widely in its characteristics as a result of it being purified from natural sources. Three of these characteristics are of
particular importance in determining the suitability and performance in a particular application: Gel strength, Melting/Gelling temperature
and Electroendosmosis (EEO) resulting from sulfate and pyruvate content. MIDSCI offers premium quality agaroses for a wide-ra n ge
of nucleic acid and protein electrophoresis applications. Due to the superior particle size of MIDSCI agarose blends, lower background
fluorescence is routinely achieved following ethidium bromide staining. All agaroses are rigorously tested to meet the highest quality control
standards and to ensure success in your molecular biology application.
